Clodronate Liposome Has Numerous Scopes

Clodronate Liposome, or clodrosome, is used for suppressing macrophage activity. Clodronate can reduce development of cancer metastases, reduce pain and risks of bone fractures, but it cannot be delivered to tissues without a little help of liposomes. This nano technology allows very effective delivery of this compound, creating macrophage depleted tissues and organs.

To be closed in vivo, this effective compound is delivered encapsulated within liposomes. A liposome is lipid-based tiny particle, designed to carry different compounds in form of suspension. It is lately used in many research laboratories, as highly effective way or delivering drugs to various tissues. In this case, to bones and kidneys.

Clodronate liposome suspension should be injected to targeted tissues. While manipulating this type of suspension, some measures of precaution are necessary, such as chemical resistant gloves and clothing, safety goggles and respirators. This compound can be harmful if inhaled, absorbed through skin, swallowed or if it get in contact with your eyes. It causes different irritations, especially to mucous membranes, and you should be very careful with it.

In case of oral exposure, you should wash out mouth with water and call a physician. If your eyes were in contact with this compound, you should flush them with water for at least fifteen minutes. Dermal exposure should be treated with soap and water. If inhaled, it may cause breathing problems. Exposed person should get to fresh air, and sometimes it might be necessary to use respirator or even oxygen.

Macrophages are very important, because they regulate immune reactions and some other, non-phagocytic cells function. Their primary function is to ingest and digest unfamiliar macromolecules and other microorganisms. In order to override their function, they use liposomal encapsulated compounds and drugs.

When liposomes encapsulated clodronate gets to tissue, macrophage will ingest and digest it, but they will only destroy this liposome capsule, not the active component. It will be released within this tissue, and then accumulated. Macrophage depleted tissue will allow performing different in vivo researches related to macrophages.

This suppresion of macrophages activity is especially useful for different in vivo researches of various models of neurological disorders and gene therapy, as well as for numerous autoimmune diseases. It is designed to be used for in vivo researches. Even so, it can also be used for some in vitro researches.

It is important to keep the suspension at appropriate temperature. It shouldn't be exposed to temperatures above 30 degrees of Celsius, or frozen. The ideal storage temperature is four degrees Celsius, and in this case it can be kept one month in refrigerator. For achieving desired results, the suspension should be kept outside the refrigerator until it reaches room temperature, before injecting it to animals.

It is also important to shake the suspension until it becomes homogeneous. You should shave the skin and cleanse it with alcohol, to make sure it is disinfected. You should use sterile syringe for each animal. If you use the same one, you might get uneven dosages. Follow the procedure for injecting clodronate liposome to your test animals.
